Fargoland

This is time-resolved fluorimeter based on direct digitizing of luminescence signal excited by short light pulse. We use second or third harmonic of passive Q-switched YAG microchip laser as excitation source and 8 Gs/sec digitizer DS252 (Acqiris, Geneva, Switzerland). This instrument requires less than second for data acquisition and lets recover information about luminescence with lifetime as short as 0.1 ns. Basically we have two instruments what share digitizer and controlled computer:
  • Stand-alone instrument based on optical table and uses optical filter for wavelengths separation. A flow-through temperature bath is coupled to the instrument, allowing control of the temperature during the measurements
  • Accessory for Eclipse fluorimeter what fits in Eclipse sample compartment and uses its emission monochromator. The accessory is equipped with Peltier temperature-controlled cuvette holder TLC 50 (Quantum Northwest, Spokane WA) with magnetic stirring and four open windows for fluorescence.
  • Instrument controlling software can carry out simple single waveform experiments or automatically create 3-D experiments with many waveforms as function of emission wavelength, emission polarizer orientation, and delay time. For delay time based experiments stand-alone instrument has stopped-flow accessory what is customized SFA-20 mixing accessory (Hi-Tech Scientific, Bradford on Avon, United Kingdom). Moreover user can manually perform series of experiments based on titration, temperature or another conditions.
    Recently stopped-flow module SFM-20 (Bio-Logic, Claix, France) was integrated in instrument. It consists of two step motor driven syringes and optical observation head. The original light collection and detection systems were designed for this accessory. This accessory can be used for double-kinetic experiment that is a simultaneous detection of luminescence changes in nanosecond and millisecond time scales. Because of efficient design it has stopped-flow dead time only about 2 ms.
    Our state-of-art data analysis software is designed to globally analyze virtually infinite number of waveforms using extremely sophisticated fitting models. Model is designed by user as a combination of processes such as light emission, quenching, resonance energy transfer, rotation molecule diffusion, chemical reactions, etc.
